Active ingredient: Propylthiouracil

Brand Name: PropylThyracil, PTU

Drug class: Antithyroidal drug

Mechanism of Action:

Propylthiouracil (PTU) is used in women to treat hyperthyroidism, including Graves’ disease and during pregnancy, where managing thyroid hormone levels is critical.

Mechanism of action: PTU works by inhibiting thyroid hormone synthesis, blocking the enzyme thyroid peroxidase, and reducing T3 and T4 production. It also inhibits the conversion of T4 to the more active T3.

Effect on fertility biomarkers

PTU doesn’t directly affect fertility, but controlling hyperthyroidism helps regulate menstrual cycles and reproductive health. Temperature effect: PTU is presumed to lower core body temperature by impairing metabolic thermogenesis. This potential decrease in body temperature may affect basal body temperature (BBT) charting, which is used for fertility tracking.
